First gene therapy study in human salivary gland shows promise

ScienceDaily (Nov. 5, 2012) ? Gene therapy can be performed safely in the human salivary gland, according to scientists at the National Institute of Dental and Craniofacial Research (NIDCR), part of the National Institutes of Health.

This finding comes from the first-ever safety, or Phase I, clinical study of gene therapy in a human salivary gland. Its results, published this week in the Proceedings of the National Academy of Sciences, also show that the transferred gene, Aquaporin-1, has great potential to help head and neck cancer survivors who battle with chronic dry mouth. Aquaporin-1 encodes a protein that naturally forms pore-like water channels in the membranes of cells to help move fluid, such as occurs when salivary gland cells secrete saliva into the mouth.

These initial results clear the way for additional gene therapy studies in the salivary glands. Although sometimes overlooked, salivary glands present an ideal target for gene therapy. They are easily accessible and, once a gene is introduced, it has no obvious escape route into the bloodstream, where it can have unintended consequences.

"You cannot imagine how fulfilling it is to jot down an idea on a napkin in 1991 and then see it enter a clinical trial and help people.," said Bruce Baum, D.M.D., Ph.D., lead author on the study and recently retired NIDCR scientist who spent the last 21 years moving gene therapy in the salivary glands from the research bench to the clinic. "Can a scientist ask for anything better?"

Baum's interest in helping head and neck cancer survivors dates to the early 1980s. While attending to patients in the NIDCR's Dry Mouth Clinic, Baum encountered numerous people with head and neck cancer who had received radiation therapy to shrink their tumors. The radiation, while effective in treating cancer, had inadvertently damaged nearby salivary glands, compromising their ability to secrete saliva into the mouth.

Baum said he was thoroughly frustrated at the time because he had no effective moisture-restoring treatments to offer most patients. They had beaten cancer, but the radiation had left them with a permanent parched sensation in their mouths that diminished their quality of life and often led to chronic oral problems, such as difficulty swallowing, inflammation, infection, bad breath, and pain.

In the early 1990s, as the first gene-therapy studies entered research clinics, Baum saw an opportunity to make a difference. An initial napkin sketch of the procedure and 15 years of research later, Baum and his colleagues had assembled a compelling scientific case in animal studies that the transferred Aquaporin-1 gene, once expressed, will create new water channels in the impermeable salivary gland cells and allow water to flow through them. After rigorous reviews by NIH and the U.S. Food and Drug Administration, the Phase I protocol was launched and the first patients treated in 2008.

The scientists gave 11 head and neck cancer survivors a single-dose injection of the Aquaporin-1 gene directly into one of their two parotid salivary glands, the largest of the major salivary glands. The gene was packaged in a disabled, non-replicating adenovirus, the cause of the common cold when intact but incapable of causing a cold in this case. As is standard in gene therapy studies, the virus served as the vector, or Trojan horse, to deliver the gene into the cells lining the salivary gland.

The scientists found that five participants had increased levels of saliva secretion, as well as a renewed sense of moisture and lubrication in their mouths, within the study's first 42 days, the period covered in this report. Of the six who didn't benefit from gene therapy, none had serious side effects. The most common side effect was a transient and relatively minor immune response against the disabled adenovirus.

"It is time to evaluate a different vector to deliver the Aquaporin-1 gene, one that will cause only a minimal immune response," said Baum. "But these data will serve as stepping stones for other scientists to improve on this first attempt in the years ahead. The future for applications of gene therapy in the salivary gland is bright. "

Democrats need stronger Illinois showing to retake U.S. House

ROMEOVILLE, Illinois (Reuters) - Democrats appear unlikely to regain a majority in the U.S. House of Representatives on Tuesday despite a nationwide push, and a major reason could be disappointment in President Barack Obama's home state.

In Illinois, Democrats began this election year determined to reverse a Republican gain of four U.S. House seats in 2010 that gave them a majority of the Illinois delegation in a state considered solidly Democratic.

The powerful speaker of the Illinois state House, Michael Madigan, used the Democratic stranglehold on state government to redraw election districts after the 2010 Census in a way that would help Democratic candidates.

The strategy worked to an extent, putting Republicans on the defensive, and making six of the 18 Illinois congressional races competitive. They have attracted more than $45 million in outside spending.

Some political analysts say Democrats could gain three seats back from Republicans in Illinois, which would probably not be enough to return the national party back to power on Capitol Hill.

"The Democrats are not going to take the House, the hill is simply too steep for them to climb," said Michael Mezey, a political science professor at DePaul University in Chicago.

Republican U.S. Rep. Joe Walsh, an incumbent backed by the conservative Tea Party movement, was widely expected to lose to Tammy Duckworth, an Iraq War veteran featured at the Democratic National Convention, in a heavily Democratic district. Two other Republican incumbents, including the relatively moderate Judy Biggert in a suburban Chicago district, are in very tight races.

Democrats also could gain five seats in California, but the rest of the national map doesn't bode well for them.

Republicans used their victories in state legislatures and governors' races in 2010 elections to redraw voting districts in states such as North Carolina to offset the expected Democratic victories elsewhere.

Mezey expects Democrats will pick up "probably no less than five seats and certainly no more than 10," nationally, well short of the 25 seats they need to return to the majority of the 435-member House of Representatives.

"A BULWARK" AGAINST NANCY PELOSI

Unlike Joe Walsh, whose reputation as a Tea Party firebrand made him vulnerable to a challenge from Tammy Duckworth even before he made controversial remarks about abortion, Biggert is running as a bipartisan moderate.

The candidate repeatedly described herself as a compromiser when she spoke recently to senior citizens at the Tasty Waffle restaurant in Romeoville, a outer suburb of Chicago where strip malls mingle with open farmland.

Ken Scorza, 68, a retired independent, said he supports Biggert because "she is one of the few people who constantly reach across the aisle for the betterment of the country."

Appearing alongside Biggert was U.S. Rep. Peter Roskam, a senior House Republican from a neighboring district, who highlighted the threat of a Democratic majority.

"Can you imagine bringing Nancy Pelosi back?" he asked, referring to the Democratic House Minority leader whose name raises conservative hackles. "The bulwark against that is to send Judy Biggert back to Congress."

Biggert's opponent, scientist and former Democratic Rep. Bill Foster, describes her as an extreme politician. At a rally in Aurora, a Chicago suburb with a large Hispanic population, Foster said Biggert "sided with the Tea Party Congress" against the Dream Act, which would have provided a path to citizenship for illegal immigrants brought to the United States as children.

"Bill Foster understands that a healthy middle class is crucial for America's economy," said Cassidy Alexander, a self-employed artist who attended the rally. "He is a scientist who makes rational decisions and that is really important to me."

That race alone has brought in $8 million in outside money, bringing a barrage of mostly negative television ads that often run back to back in the suburbs.

Obama won Biggert's strangely shaped district easily in 2008, making it more difficult for her to hold onto the seat.

Another tight race involves Republican Robert Dold, a freshman congressman from Chicago's North Shore, whose district was altered to include less of the Republican northern suburbs and more ethnically mixed neighborhoods nearer to Chicago. His race has attracted nearly $7.3 million in outside money.

DePaul's Mezey said Dold has a solid chance of fending off Democratic challenger Brad Schneider.

"If Dold does win, he'd have the singular honor of being the Republican in the House with the most Democratic district in the country," he said.

(AP) ? France's president pledged Sunday that his country will stand against instability in Lebanon, two weeks after the assassination of a senior Lebanese intelligence official sparked clashes between pro- and anti-Syrian groups.

Later in Saudi Arabia, Francois Hollande held further talks on Syria with King Abdullah, whose country has been a leading supporter of the forces trying to overthrow Syria's President Bashar Assad.

In Beirut, Hollande promised that Paris and the European Union will help Lebanon deal with an influx of more than 100,000 refugees who have fled the civil war in neighboring Syria.

"We are committed to give you guarantees regarding security, stability and unity of Lebanon," Hollande told reporters after meeting President Michel Suleiman.

The Oct. 19 car bomb that killed Brig. Gen. Wissam al-Hassan, a powerful anti-Syrian intelligence official, stirred up deadly sectarian tensions in Lebanon, where Sunnis and Shiites are deeply divided over the civil war in Syria, which has killed at least 36,000 people since it began in March 2011.

Lebanon's two largest political coalitions have lined up on opposite sides of the conflict. The powerful Shiite group Hezbollah and its partners who dominate the Lebanese government have stood by Assad, while Lebanon's Sunni-led opposition backs the rebels seeking to topple the Damascus regime.

Assad and many in his inner circle are Alawites ? an offshoot of Shiite Islam and a minority in Syria ? while the rebels come mostly from the country's Sunni majority.

Syria dominated Lebanon for 29 years after it first sent troops into its smaller neighbor in 1976, during Lebanon's 15-year civil war.

Damascus' three-decade hold on Lebanon began to slip in 2005, after the assassination of former Lebanese Prime Minister Rafik Hariri. Still, for years after Syrian troops pulled out, there were frequent assassinations of anti-Syrian figures in Lebanon, and the perpetrators have yet to be tracked down.

Despite the Syrian military's withdrawal from Lebanon, Assad has managed to maintain his influence in the country through allies, such as Hezbollah.

Hollande made a three-hour stop in Beirut en route to Saudi Arabia for talks that focused on the Syria conflict and Iran's disputed nuclear program. Saudi officials said Hollande arrived in the Red Sea port of Jiddah, accompanied by a high-level delegation that included French Foreign Minister Laurent Fabius and a senior trade official.

France's ambassador to Saudi Arabia said in comments published in the Al-Riyadh newspaper Sunday that Paris and Riyadh share views on both issues. The newspaper quoted Bertrand Besancenot as saying France backs tighter sanctions on Iran and calls on Syrian President Bashar Assad to step down.

Saudi Arabia is Iran's main regional rival and is a key backer of Syria's rebels.

While in Beirut, Hollande said France will give "full assistance" in the investigation of al-Hassan's assassination, saying "the Lebanese and the world want this. There will be no escape from justice."

Al-Hassan was killed a day after he returned home from Paris, where his family has been living for years.

"Even though Lebanon is very close, it should not be the victim of this crisis," Hollande said referring to Syria's civil war. "We renew France's full support to the stability, unity, independence and safety of the territory of Lebanon."

France, the onetime colonial ruler of both Syria and Lebanon, has been one of the most outspoken Western critics of the Assad regime. France announced in September that it had begun sending direct aid and money to five rebel-held Syrian cities as part of its intensified efforts to weaken Assad. It was the first such move by a Western power amid mounting calls for the international community to do more to prevent bloodshed.

French officials have acknowledged providing communications and other non-lethal equipment to Syrian rebel forces, but say they won't provide weapons without international agreement. Paris played a leading role in the international campaign against Libyan dictator Moammar Gadhafi last year.

France still wields wide influence in Lebanon and has about 900 peacekeepers deployed near the border with Israel.

I think one of the problems readers faced is that when viewed the list on my tracker, they will still need to go to SGX.com to find out the frequency of distribution.

The frequency can be pretty consistent, but it needs manual updates on my end. But I will work out a schedule to keep it up to date weekly.

Acupuncture relieves fatigue in breast cancer sufferers

(NaturalNews) If you or someone you know suffers from breast cancer and the chronic fatigue symptoms commonly associated with conventional treatment protocols, acupuncture just might provide the much-needed energy boost you need to make it through each day. Researchers from the University of Manchester in the U.K. found that routine acupuncture treatments can help women with breast cancer achieve a "mild to modest" improvement in their fatigue symptoms, which for some can make all the difference in determining overall quality of life.

For their research, Alexander Molassiotis and his colleagues monitored and tracked 302 women with moderate to severe cancer-related fatigue symptoms, all of whom had received chemotherapy treatments for their breast cancers in the five years preceding the study. A total of 227 women who participated in the study received acupuncture treatments, while the remaining 27 women did not receive the treatments. After six weeks, the team compared the health conditions of the two groups to see if acupuncture had provided any benefits.

The group found that, overall, the acupuncture group experienced a drop in fatigue symptoms of almost four points on a 20-point scale, compared to a less than one point drop in the non-acupuncture group. Anxiety and depression scores also improved among those in the acupuncture group, dropping by two additional points following acupuncture compared to women given educational materials only. Emotional and physical well-being also improved as a result of acupuncture, lending credence to the notion that acupuncture is capable of transforming both mind and body for the better.

"Acupuncture is an effective intervention for managing the symptom of chemo-related fatigue and improving patients' quality of life," wrote Molassiotis and his colleagues in their report. When combined with moderate exercise, talk therapy, and other physical and emotional exercises, acupuncture can play a crucial role in mitigating the fatigue-related side effects of breast cancer treatments.

A 2008 study published in The International Journal of Radiation Oncology found that acupuncture is also effective at treating a variety of other side effects of cancer treatments as well, including hot flashes, depression, and night sweats. Foursquare Looks Into An Fourth Round At An Over $700M Valuation, Investors Skeptical

In previous rounds, the company has brought in over $71.4 million in financing from Andreessen Horowitz, Spark Capital, Union Square Ventures and others. From what we're hearing the company seeks to flesh out this round at a pre-money valuation of between $700 million and $800 million, and that number has a few VC shops skeptical due to its growth trends, which have not gotten more inspiring after the company's?recent June redesign, according to multiple sources. Foursquare raised a?$50 million round at a $600 million valuation in June of 2011 and still has plenty of money in the bank as far as we know.
